Is it possible in Artisteer to set up a different layout for the homepage than for the rest of the pages? I want the homepage to have a single horizontal menu under the header with a single column under it and the rest of the pages to have the same menu but with a 3-column body.

Artisteer doesn't support selecting unique layout for every page, but you can do it using additional page templates if you are using Wordpress.

A CMS? Yes.
If you're doing it as a HTML then you just need to delete the relative DIVs.
What are you doing this in?

This is HTML so I guess I'll just build out the template I want and then hollow out the first page.

It is easy to do with Joomla also.

Yes in case of HTML you need to do it manually after exporting the site from Artisteer.
